将香榧(Torreya grandis‘Merrillii’)的未成熟合子胚置于SH+0.1 mg·L-1 NAA+500 mg·L-1AC+3%蔗糖+0.5 g·L-1 Gln培养基上暗培养45 d,诱导产生半透明颗粒状胚性愈伤组织;将胚性愈伤组织转入SH+20 g·L-1 PEG+10 mg·L-1 ABA培养基中暗培养3个月诱导体细胞胚。采用碘—碘化钾染色和石蜡切片技术对体胚起源、形态发育与细胞组织学进行了观察。结果表明:胚性愈伤组织起源于合子胚胚轴表皮或皮层细胞的对称分裂。胚性愈伤组织含有两类细胞,一种是细胞质浓厚、细胞核大、体积小的圆形胚性细胞,另一种是高度液泡化拉长的细胞。胚性愈伤组织包含由这两种细胞构成的原胚团Ⅰ、原胚团Ⅱ和原胚团Ⅲ,以及一些游离细胞。原胚团Ⅲ在无植物生长调节剂的SH基本培养基上形成原胚,原胚接入成熟培养基,历经球形、棒状、心形、鱼雷形胚后发育成子叶胚。将子叶胚转入萌发培养基后胚根伸长,胚芽发育长出针叶,形成完整的再生植株。同时在离体培养中合子胚胚柄处退化的裂生多胚也能重新发育形成胚体。在初生体胚发育过程中表面常伴有次生体细胞胚的形成。 相似文献

就贵州省赤水市料慈竹(Bambusadistegus)0~4a5个龄级的纤维形态进行显微观测和系统分析表明,其纤维长度为2.13~2.69mm(平均2.45mm),纤维宽度为13.42~15.06μm(平均14.09μm),长宽比历8~185(平均174),壁厚2.52~6.46μm平均4.86μm),腔径2.15~8.39μm(平均4.41μm),壁腔比为0.6~6.17(平均3.24)。通过方差分析后表明,除纤维长度和宽度外,年龄对料慈竹纤维形态有一定影响,表现在纤维壁厚度上,从0~1a有增厚,1a后相对稳定,至4a时明显增厚;纤维腔径相反,从0~1a腔径减小,至4a时明显减小。 相似文献

借助光学显微镜和扫描电镜研究了碱蓬属2种植物碱蓬和盐地碱蓬的花粉形态,并对不同生境条件下盐地碱蓬4变式的花粉形态进行了对比研究,研究结果表明:花粉粒外部形态成球形;大小在15.4-24.0μm之间;具有散萌发孔,萌发孔数量在46-55之间,萌发孔直径为3.1-5.4μm;花粉粒表面光滑,其上散布许多颗粒状小突起,萌发孔上亦有数目不等的小突起。以上数值在不同种间有较明显的差异,在盐地碱医宵同生态变式间亦有差异,但差异较小。这些细微的特性差异可为碱蓬属植物分类提供参考依据,是研究该属植物分类及生态的新资料。 相似文献

To elucidate the differences in the leaf water relations of Pinus densiflora Sieb. et Zucc. growing in different soil moisture conditions, we examined the pressure-volume curve and the diurnal changes in the stomatal conductance, the transpiration rate, and the leaf water potential. The leaf water relations were compared using field-grown 40-year-old pine trees growing on the upper and lower parts of a slope. We also compared the leaf water relations of potted 4-year-old saplings growing at pF 4.2 and pF 1.8 soil moisture levels for almost 1 year. The values of the ratio of symplasmic water at turgor loss point to symplasmic water at saturated point (Vp/Vo) and bulk modulus of elasticity () of both the adult trees on the upper part of the slope and the potted saplings growing on pF 4.2 soil moisture were higher than those values of both the adult trees on the lower part of the slope and the potted saplings growing on pF 1.8 soil moisture, respectively. The field-grown adult tree and the potted saplings growing under long-term water stress tended to reduce their stomatal conductance in response to the acute soil drying. It is suggested that P. densiflora growing under long-term water stress rapidly closed its stomata in response to soil drying and avoided losing water, and could also rapidly absorb water with reducing water loss because of the decrease in the leaf pressure potential derived from the high values. 相似文献

松材线虫病是一种毁灭性的森林病害,不同地区的松材线虫在形态特征方面有一定的差异.采用扫描电子显微镜对来自湖南、广东两地3个株系松材线虫进行了形态观察比较.结果表明:这3个株系的松材线虫成虫的头部、尾部形态特征无明显差异,幼虫和成虫的唇区与体部都出现缢缩;松材线虫口针的开口端呈一斜面;雄性成虫尾部交合伞呈“M”型.这些特征都是在过去的研究中没有描述过的,可为松材线虫的形态研究提供参考依据. 相似文献

[Objective]To compare the differences of seed morphological characters among provinces of Quercus variabilis Bl.,and explore the adaptive growth status of seedlings growing in the central distribution area. [Method]The seeds of 6 Q. variabilis provenances from six provinceswerecollected, and the seedlings were cultivated and transplanted to experimental field in the centre of natural distribution range (Wuhan Jiufeng National Forest Park), then the seed morphology and one-year seedling growth traitswere surveyed. [Result](1)There were significantor extremely significantdifferences among provenances in seed length, seed width, seed length/width ratio and 100-seed mass, the order of seed length among provenances was Beijing Provenance (22.28 mm),Hubei Provenance (22.10 mm), He'nan Provenance (20.80 mm), Shaanxi Provenance (19.57 mm), Yunnan Provenance (18.93 mm)and Liaoning Provenance (18.81 mm), the seed width of Beijing Provenance (19.71 mm) and Hubei Provenance (19.16 mm) was higher than that of the others, the seed length/width ratio of He'nan Provenance (1.38) was extremely significantly higher than that of the others; (2)There were significant or extremely significant differences among provenances in seedling height, ground diameter, leaf number, branching number and seedling height/ground diameter, the Beijing Provenance and Hubei Provenance were better in seedling height and leaf number, which were respectively 19.54 cm and 20.26 cm, 8.32 leaves and 9.50 leaves; (3)A decreasing trend with time for seedling growth traits among provenances was found; (4)There was a positive relationship between seed morphological characters and seedling growth traits except seed length/width ratio; the leaf number was significantly related to ground diameter and branching number and the seedling height and leaf number were closely related to the seed morphological characters; (5)It showed a decreasing trend with the growth time in the relationship between seedling growth traits and 100-seed mass. [Conclusion]The seedlings of Beijing Provenance and Hubei Provenance grew better in the experimental field which might be related to their larger seed mass, and the Yunnan Provenance in the south margin and the Liaoning Provenance in the north margin of distribution area of Q. variabilis grew weaker than the provenances in the centre of distribution area.With the effect of climate change, provenances in the margin of distribution area may lose in the community competition in the seedling stage if it cannot take advantage of its dispersal and settlement and affected by the disadvantage of growth and competition, so that the distribution and population renewal would surely be affected. 相似文献

通过对毛竹材纤维形态,组织比量、纤维素含量和基本的分析测定,研究了不同年龄、不同胸径、竹秆不同部位栲生性状的变异特点结果表明,竹材纤维长度主要受竹秆部位的影响,与年龄和胸径无显著相关,组织比量与年龄无显著相关,与胸径有一定相关;纤维素含量和基本密度与年龄、胸径、竹秆部位均有关系,从材性变异特点来看,经营培育小径级毛竹对制浆造纸更有利。 相似文献
